She also took classes in horse riding while working at Jayaprabha Studio. After that, she went to Pune, where she started working with Mangal Pictures. In 1953, she shifted to Bombay from Pune. Sulochana Latkar marked her debut in films in 1946. From 1946 to 1961, she worked as a lead actress in Marathi films such as "Sasurvas” (1946), “Sangtye Aika” (1959), “Bhaubheej,” “Mothi Manse,” “Lakshmi Ali Ghara,” “Jivacha Sakha,” Sukhache Sobti,” “Akashganga,” and “Dhakti Jau.” Throughout her career in Hindi films, Sulochana Latkar was often paired opposite Trilok Kapoor, Nazir Hussain, and Ashok Kumar. She once said in an interview that she loved portraying the mother role to three actors Dev Anand, Rajesh Khanna, and Sunil Dutta.

She is often portrayed mother or as a close relative in Hidi-language films with Sunil Dutta, such as Jhoola, Heera, Sujatha, Mehrbaan (1967), Ek Phool Char Kante, Bhai Bahen (1969), Reshma Aur Shera, Umar Qaid, Jaani Dushman, and Badle Ki Aag. She worked regularly in films with Dev Anand in the lead roles in such films, as Jab yar Kisise Hota Hai, Duniya (1968), Pyar Mohabbat, Jhonny Mera Naam, Amir Garib, and many more. She is often portrayed on screen close relative to the character played by Rajesh Khanna in Dil Daulat Duniya, Mere Jeevan Saathi, Prem Nagar, Adhikar (1986), Aashiq Hoon Baharaon Ka, and many more. Sulochana Latkar was a recipient of India's fourth-highest civilian award, the Padma Shri, in 1999.

In 2003, she was honored with the Chitrabhushan Award. In 2004, she earned the Filmfare Lifetime Achievement Award. She won the Hridaynath Mangeshkar Award four times. In 2009, she earned Maharashtra’s highest civilian award, Maharashtra Bhushan by the Government of Maharashtra. In 2019, she was awarded with Saraswatibai Dadasaheb Phalke Lifetime Achievement Award. Sulochana Latkar died on June 4, 2023, in Dadar in Mumbai, Maharashtra. She died due to respiratory failure at Shushrusha Hospital.
