Together, they formed a pioneering theater group named ‘Bohurupee’ in 1948 and produced many Tagore plays on stage. She made her debut on the stage in her cousin Bijon Bhattacharya's play Agun or 'Fire' in 1943. She also acted in Bhattacharya’s production’ Jabanbandi’ or ‘Nabanna’ in the following year. She got married in 1945 and quit Indian People Theater Association in 1947 and formed her own theater group ‘Bohurupee’. She also acted in Hindi movies essaying low-caste rural heroine in Mahesh Kaul's Gopinath (1948) and played the hero's widowed mother in Khwaja Ahmad Abbas's ‘ Munna Click to look into! >> Read More... ’ (1954). But her reputation rose with her roles in the production of Tagore’s plays.

She played substance characters in Tagore’s ‘ Char Adhyay’(1951) and ‘Raktakarabi’ (1854). She brought a new drama movement in Bengal theater and her acting and stage presence arrested spectators for her deep understanding of characters, her expressive face always reflected the subtle mental states fo people, and her astonishing use of voice established a good rapport with viewers. Tripti Mitra once stated that she never thought acting in the theater would become a career for her as she only wanted to become a doctor or a nurse. Her first Bengali film was Pathik in 1953; the film was directed by Debaki Kumar Basu. She also acted in Ritwik Ghatak's last film, Jukti Takko Aar Gappo (1974). She also acted in Urdu movie ‘Jago Hua Savera’(1959), which was produced in Bangladesh and was an adaptation of a story based on Manik Bandyopadhyay's classic novel "Padma Nadir Majhi".

Sombhu and Tripti had a daughter, Shaoli, who is also a noted actor, director, and playwright. Sombhu Mitra and Tripti acted together in Bengali films too.
