Tamhankar was born on June 25, 1986, to parents Nandkumar Tamhankar and Mrunali Tambadkar. Her mother, Mrunali is also an actress known chiefly for the Marathi films Party and Bonus.

Born in Sangli, Maharashtra, Sai schooled at Savarkar Pratishthan School in Sangli. As a teenager, the actress played Kabaddi at the state level and secured an orange belt in Karate. Sai pursued graduation at Chintaman College of Commerce in Sangli.

Tamhankar’s interest in acting was ignited when she performed in a play helmed by her mother’s friend. Later, she participated in a play titled Aadhe Adhure, which led to her winning the Best Actress title at an Inter-College competition. Although Tamhankar has featured in many movies and shows, the actress shot to fame with her performance in Hya Gojirvanya Gharat. Tamhankar bagged the Filmfare Award Marathi for Best Supporting Actress for Family Katta in 2018. She has to her credit other awards like Best Actress 2015- Newsmakers Achievers, Maharashtra Achievers Award 2018 for Entertainer of the Year, and many more.

She is the first Marathi actress to grace the cover of Femina Magazine twice in 2015. Tamhankar also owns a wrestling team called Kolhapuri Mavle for the tournament Zee Maharashtra Kusti Dangal.
